Perspective: Harnessing the Mechanism of Melanin Radiotrophy for Energy Harvesting and Radiation Protection in Peripheral AI-Symbiont (PAIS) — Implications for Deep-Space Survival and Upstream Intervention in Aging
Abstract
Radiotrophic fungi, particularly melanized species such as Cladosporium sphaerospermum, demonstrate a remarkable ability to utilize ionizing radiation as a supplementary energy source through melanin-mediated radiosynthesis. This Perspective proposes integrating this natural mechanism into the architecture of the Peripheral AI-Symbiont (PAIS) — a fully peripheral network of self-organizing micro/nano-robots designed for upstream intervention in inflammaging via disruption of the Peripheral Vicious Cycle (PVC). By engineering thin polydopamine (PDA) coatings — synthetic melanin analogs — onto PAIS agents, a dual-function layer is introduced that provides passive radioprotection and active energy harvesting under high-radiation conditions. In physiological settings, the coating acts as an antioxidant and mechanical stabilizer. Under extreme radiation (deep space, radiotherapy, or age-related mitochondrial decline), it switches to radiosynthesis mode, converting ionizing radiation into usable chemical energy. This backup pathway enables sustained PAIS operation even when host cellular metabolism is compromised, supporting continuous monitoring, micro-vibration delivery, matrix repair, and anti-inflammatory activity in peripheral tissues. The approach preserves PAIS core principles of peripherality and central nervous system safety while dramatically enhancing system autonomy and resilience. Structural integration, operational modes, synergy with existing anti-aging functions, potential challenges, and implementation pathways are discussed. If realized, melanin-augmented PAIS could become a cornerstone technology for both long-duration interplanetary missions and radical healthy longevity interventions on Earth, turning a major space hazard into a renewable resource for tissue maintenance and repair.
