Abstract

Radical human lifespan extension faces two coupled biological barriers: telomere attrition (replicative senescence) and genomic instability (oncogenesis). Previous therapeutic attempts have failed due to vector immunogenicity or the carcinogenic risks of telomerase activation. This paper presents the "Bergel Unified Protocol," a six-dimensional engineering framework designed to overcome these hurdles. The protocol integrates: (1) Synergistic Payload: hTERT activation coupled with Whale-derived Super-p53; (2) Advanced Stealth Technology: CD47-enriched Exosome-AAVs to bypass phagocytosis; (3) Active Safety Systems: miRNA-detargeting for liver protection and an iCasp9 "Kill Switch"; and (4) Aseptic Clinical Isolation. Furthermore, we propose the use of Epigenetic Clocks (Horvath) as the primary biomarker for efficacy. Stochastic Monte Carlo simulations demonstrate a theoretical survival extension to >140 years with minimized oncogenic risk.
