Unravelling the Mechanisms of Partial Reprogramming: Chromatin Governance, Transcription Factor Redistribution and Ageing
Abstract
Abstract Partial epigenetic reprogramming can restore youthful molecular and functional characteristics while preserving differentiated cellular identity, yet the mechanisms responsible for these effects remain incompletely understood. This hypothesis-driven perspective integrates recent advances in pioneer transcription factor biology, nucleosome geometry, chromatin architecture, developmental regulation and epigenetic network organisation to propose a broader mechanistic interpretation of partial reprogramming. Rather than viewing rejuvenation primarily as reversal of individual molecular ageing markers, this Perspective suggests that transient expression of pioneer transcription factors may temporarily restore the precision of chromatin-mediated regulatory organisation while preserving cellular identity. The manuscript further explores the distinction between biological ageing and survivability, and considers how imbalance between regulatory plasticity and stability may contribute to both rejuvenation and disease.
