Abstract

We describe a function for the TIR domain TLR/IL-1 family receptor SIGIRR (TIR8) in clearance of transforming, transformed, dead and dying cells at the plasma membrane through a mechanism involving recruitment of immune cells. The mechanism likely centered on a process featuring membrane dynamics governed by LLGL2 and TMBIM6 and involved the function of the cadherin-like CDHR5 in the context of oncogenic-type signaling by MAP3K11, APRT and CDK10. SIGIRR likely functioned in concert with a second receptor recently described in coupling of developmental processes with resemblance to transformation to an immune clearance process, FCGRT, as well as the PRR family receptor PRR13.
