Klotho and the Two-Layer Model of Kidney Essence: A Molecular Extension of the OMN™ Aging Hypothesis
Abstract
Oriental Medicine Neurodynamics (OMN™) is a theoretical framework that reinterprets the classical East Asian medical concept of "kidney essence" (shen jing) in terms of autonomic nervous system functional dynamics. A prior paper on aging (Ishihara, 2026) proposed that the rate at which kidney essence (yin qi / water) is depleted determines the rate of biological aging, drawing on clinical observation and Rattan's homeodynamics/hormesis framework. The present paper extends this hypothesis by proposing that kidney essence can be decomposed, on embryological grounds, into two distinct layers: (1) a Ministerial Fire system, mobilized on command from the Heart–Wood (sympathetic) axis and originating in the adrenal gland, and (2) a Kidney-Essence-Proper system, autonomously regulated by the kidney (nephron) itself in response to internal physiological state. We further propose klotho—a circulating hormone produced in the renal tubules that declines consistently with age—as a candidate molecular substrate for the Kidney- Essence-Proper system. The physiological profile of klotho (Kuro-o et al., 1997; Kurosu et al., 2005; Maltese and Karalliedde, 2012; Prud'homme et al., 2022) converges, at the molecular level, with both the classical and OMN™ propositions that kidney essence is an innate reserve endowed at birth that declines progressively with age. The two-layer model presented here opens the possibility of eventually discussing individual variation in aging rate in terms of "the rate at which excessive mobilization of the Ministerial Fire system depletes the Kidney-Essence-Proper system," and is presented at this stage as a clinical hypothesis (Tier 2/3).
