Abstract

Expanding the templated-editing toolbox, here we describe THOR (Template Homology Oriented Repair). THOR is a CRISPR-Cas9-based technology that engages endogenous DNA-dependent polymerases through DNA-tailed guides to enable precise editing without an exogenous polymerase, which can simplify editor synthesis and delivery. We demonstrated that THOR enables the installation of a variety of genome edits within multiple genomic targets, cell models, and delivery modalities.
