Organ aging measures have varied genetic links to disease

Clinical Epigenetics

In European-ancestry genetic datasets, researchers analysed nine UK Biobank-derived organ-specific biological age gaps and 19 age-related diseases or related traits. These gaps are model-based measures of aging-related variation. They used bidirectional two-sample Mendelian randomization, which uses genetic differences to assess associations between traits in both directions.

Links included kidney age gaps with a measure of kidney filtration, and heart-and-blood-vessel age gaps with disease of the arteries supplying the heart and stroke affecting large arteries. Heart-and-blood-vessel, metabolic and lung age gaps were linked to type 2 diabetes. In reverse analyses, genetic susceptibility to type 2 diabetes was linked to age gaps including those for the brain and liver. Patterns of shared genetics varied across individual DNA regions and across DNA as a whole. Analyses accounting for multiple traits retained only some associations.

Why it matters

The work addressed whether organ-level aging and age-related diseases fit one shared aging pattern or require a more organ-specific understanding.

Caveats

The analysis relied on genetic summary data, with differing ability to detect associations and widespread variation between estimates from different genetic variants. These limitations argue against interpreting the links as simple causal pathways.
