Common variant-based genome editing to address the heterogeneity of pathogenic variants in dominant disorders

Abstract

There are approximately 2,000 known autosomal dominant disorders, which collectively represent a substantial medical burden. CRISPR-Cas9–based genome editing offers a promising route toward disease-modifying therapies that correct or disrupt pathogenic alleles.
