Jiannao Bushen Oral Liquid (Jiannao Bushen Oral Liquid) ameliorates cognitive function in Alzheimer’s disease mice by suppressing oxidative stress via the cGAS/STING pathway
By ZHU Q, YANG M, Liu W et al.
OBJECTIVE: To investigate the inhibition of oxidative stress by Jiannao Bushen Oral Liquid (JNBSL) via the cGAS/STING pathway to improve cognitive function in Alzheimer's disease (AD) model mice. METHODS: Thirty 6-month-old male SAMP8 (senescence-accelerated) mice were randomly assigned to a model group, a donepezil group (0.65 mg/kg), and a JNBSL group (0.9 g/kg), with 10 age-matched SAMR1 (senescence-resistant) mice serving as controls, followed by continuous administration by gavage for 90 d. Cognitive function and exploratory behavior were assessed using the Morris water maze and open field tests; brain Aβ deposition and cGAS protein expression were assessed using immunofluorescence; hippocampal mitochondrial morphology was observed using transmission electron microscopy; cGAS/STING protein expression was assessed using Western blot; and levels of GSH-Px, SOD, MDA, CAT, 8-OHdG, and 3-NT in hippocampal tissue were measured. RESULTS: Compared with the model group, mice in the JNBSL group showed improved cognitive function (P<0.05, P<0.01), reduced hippocampal Aβ deposition (P<0.01), improved mitochondrial morphology, decreased cGAS/STING protein expression (P<0.0001), increased GSH-Px, SOD, and CAT activities (P<0.05), and decreased MDA, 8-OHdG, and 3-NT levels (P<0.05, P<0.01). CONCLUSIONS: Jiannao Bushen Oral Liquid can inhibit oxidative stress via the cGAS/STING pathway, delay AD-like pathological damage, and improve cognitive ability in AD mice.
