Comprehensive quality control of 3D-bioprinted constructs for skin regeneration
By Багаліка А, Луценко Т
Modern treatment of chronic wounds and other serious pathophysiological conditions of the skin is rapidly shifting from traditional treatment methods to the use of advanced therapy medicinal products. One such innovative technology is a hydrogel structure loaded with mesenchymal stem cells or products of their activity (the secretome and extracellular vesicles). Owing to their high hydrophilicity, controlled three-dimensional porous structure, and ability to mimic the architecture of the native extracellular matrix, hydrogels are recognized as ideal for creating a microenvironment that supports the metabolism, viability, and proliferation of cell cultures. The use of МСК in three-dimensional matrices and/or transdermal patches has demonstrated impressive efficacy in treating a range of pathologies that were previously difficult to treat. These pathologies include chronic diabetic ulcers, deep burn wounds, atopic dermatitis, and others. МСК function not only as building material for replacing damaged tissues but also, much more importantly, as regulators of the immune response through their immunomodulatory properties. However, despite the therapeutic potential, the transition from successful laboratory developments in vitro and in vivo to large-scale clinical application requires compliance with GLP and GMP requirements, which in turn calls for strict quality control of both finished products and individual components at every stage of the product life cycle. The aim of this review is to provide a comprehensive analysis of current methods for quality control of hydrogel constructs containing МСК, taking into account the physicochemical and biological characteristics of the system, as well as the technological and regulatory aspects that form the foundation of regenerative medicine and tissue engineering.
